Hydrogen-bonding asymmetric metal catalysis with alpha-amino acids: a simple and tunable approach to high enantioinduction.

چکیده
While asymmetric transition-metal catalysis has become a powerful method for constructing chiral products, a challenge in this field is the identification of the correct ligand for high selectivity. We report here a simple approach to chiral catalyst formation: coupling of an available pool of Brønsted acids, namely, amino acid derivatives, with tunable ligands on copper catalysts. This system can be used to generate many different chiral environments simply by changing the amino acid or ligand employed and provides a scaffold for rapid screening and identification of the correct combination for high enantioselectivity. The latter is illustrated in the copper-catalyzed alkynylation of imines in up to 99% ee.

Enantioselective Diels-Alder reactions catalyzed by hydrogen bonding.
Like molecules of life (e.g., proteins and DNA), many pharmaceutical drugs are also asymmetric (chiral); they are not superimposable on their mirror images. One mirror image form (enantiomer) of a drug can have desirable activity, the other not. Consequently, the development of methods for the selective synthesis of one enantiomer is of great scientific and economic importance. We report here t...
متن کاملA highly enantioselective amino acid-catalyzed route to functionalized alpha-amino acids.
The development of syntheses providing enantiomerically pure alpha-amino acids has intrigued generations of chemists and been the subject of intense research. This report describes a general approach to functionalized alpha-amino acids based on catalytic asymmetric synthesis. Proline catalyzed Mannich-type reactions of N-PMP-protected alpha-imino ethyl glyoxylate with a variety of unmodified ke...

The search for new stationary phases has been one of the predominant concerns in high performance liquid chromatography (HPLC) in order to achieve better resolutions, longer column lives, and reduce the time of analysis. A chromatographic packing for separation of underivatized amino acids (AAs) were prepared by dynamically coating 2-amino tetraphenyl prophyrin (atpp) on a C-18 reversed-pha...
متن کاملNew Concepts for Catalysis
Organocatalysis, the catalysis with low-molecular weight catalysts where a metal is not part of the catalytic principle, can be as efficient and selective as metalor biocatalysis. Important discoveries in this area include novel Lewis base-catalyzed enantioselective processes and, more recently, simple Brønsted acid organocatalysts that rival the efficiency of traditional metal-based asymmetric...
متن کاملCooperative transition-metal and chiral Brønsted acid catalysis: enantioselective hydrogenation of imines to form amines.
Chiral amines are an integral part of numerous important bioactive compounds. This privileged structural motif is found in naturally occurring alkaloids and amino acid derivatives as well as in pharmaceuticals, herbicides, and insecticides. Typically, enantiomerically pure amines are key building blocks in the drug-discovery process, but they are also sold on a multi-hundred-ton-scale as kineti...
